Then fill the rack with the tubes so they are ready for inoculations. Fill each tube with your desired cultures and label them accordingly. The tubes have a designated area for labeling, and you can also use a label maker for a more durable and permanent label. Post inoculation reapply the lid and tighten fully, then loosen the lid back a quarter turn to allow for gas exchange. Allow the cultures to colonize the tube for 1-5 weeks with proper gas exchange. Upon full colonization repeat assembly procedure and wrap your tubes around the lid to provide a tight seal and barrier for the lid.
